Project overview:
The goal is to develop and optimize a new therapy for patients with pancreatic ductal adenocarcinoma and the project will identify and develop new drug candidates for the treatment of pancreatic cancers. The UCF team will investigate new polyamine transport inhibitors as well as the regulation of genes associated with polyamine transport in human pancreatic cancers.

The postdoc will:
a) use CRISPR technology to generate stable cell lines with varying levels (knockout vs overexpression) of two polyamine transport genes (ATP13A3 and SLC12A8);
b) use qrt-PCR and Western blots to evaluate the degree of full length and truncated mRNA and protein levels of two polyamine transport proteins (ATP13a3 and SLC12A8);
c) determine the polyamine uptake kinetics (Vmax and Km values) of 3H-Spermidine in the new cell lines in the presence and absence of polyamine stimuli;
d) determine the performance of the new transport inhibitors using two orthogonal polyamine transport assays (DFMO/Ant44) in the new cell lines;
e) perform confocal microscopy studies to look at cell localization changes of key polyamine transport proteins (ATP13A3 and SLC12A8) in each cell line in the presence and absence of exogenous polyamines;
f) train graduate students in molecular biology techniques;
g) generate semester reports for inclusion in the grant yearly reports and;
h) write manuscripts and grant proposals related to the project.

 

Position Minimum Qualifications

For the chemistry position, a Ph.D. in Organic Chemistry, Biochemistry or equivalent from an accredited institution.

For the molecular biology position, a Ph.D. in Molecular Biology or Biomedical Sciences or equivalent from an accredited institution.

 

Preferences

The preferred candidate for the chemistry position will have experience with organic synthesis and conducting biochemical assays, as well as expertise in TLC, HPLC, chromatography, NMR, LC-MS, optical rotation measurements, column chromatography, confocal microscopy, drug design and molecular modeling experience with PyMol or equivalent software.

The preferred candidate for the molecular biology position will have experience with cell culture, protein isolation, Western blot, RNA isolation, qrt PCR, DNA sequencing, CRISPR transfections, ELISA assays, and confocal microscopy techniques.
